三氧化二***体外诱导口腔鳞癌细胞凋亡的研究

提要: 目的探讨三氧化二***(As2O3) 对口腔鳞癌细胞的体外生物学作用及机制。方法采用形态学、MTT、克隆形
成试验、DNA 梯度降解试验、原位末端标记及流式细胞仪等方法,对 As2O3 在 Tca8113 细胞系中的作用进行体外研究。结
果 As2O3 对 Tca8113 细胞产生明显的生长抑制作用,MTT显示生长抑制率为 76. 3 %。DNA 梯度降解试验、原位末端标记、
流式细胞仪显示 As2O3 诱导了 Tca8113 细胞凋亡。流式细胞仪显示 As2O3 的 Tca8113 细胞凋亡与细胞周期阻滞有关。结论
As2O3 对口腔鳞癌细胞具有显著的体外生长抑制及凋亡诱导作用。

Abstract : Objective To investigate the biological effects of As2O3 and its mechanism on human oral squamous
carcinoma cell line in vitro. Methods The effects of As2O3 on Tca8113 cell line were investigated in vitro with
morphological method , MTT , clone forming , DNA ladder , TUNEL assay and flow cytometry( FCM) . Re sults
