1 再生障碍性贫血血管内皮生长因子表达及其意义的研究作者:季健玲,刘红,孙超,姜胜华,丁润生【摘要】本研究目的是探讨再生障碍性贫血骨髓血管内皮生长因子(VEGF) 的表达情况,采用逆转录聚合酶链反应( RT-PCR )法半定量检测 7 例再生障碍性贫血患者和 12例正常人骨髓单个核细胞 VEGF 基因的表达; 用免疫组织化学法检测20 例再生障碍性贫血患者和20 例正常人骨髓组织中 VEGF 的表达。结果表明, 7 例再生障碍性贫血患者骨髓标本有 2 例表达 VEGF 基因, 表达率为 %,而 12 份正常人骨髓标本有 10 例表达 VEGF 基因,表达率为 %;再生障碍性贫血患者 VEG F 基因表达率和表达强度都显著低于正常人,两者差异具有统计学意义( ) ;免疫组织化学检测发现,再生障碍性贫血患者骨髓活检组织无 1 例表达 VEGF , 而正常人表达率为 25%( 5/20 ), 再生障碍性贫血患者骨髓活检组织 VEGF 蛋白表达阳性率低于正常人( )。结论: 再生障碍性贫血患者骨髓 VEGF 在基因转录水平和蛋白质表达水平都有显著降低, 这可能与再生障碍性贫血患 2 者骨髓血管生成减少和造血减少有一定关系。【关键词】再生障碍性贫血 Expression of Vascular Endothelial Growth Factor in Patients with Aplastic Anemia and Its Significance Abstract This study was aimed to investigate the expression of vascular endothelial growth factor ( VEGF ) in patients with aplastic anemia. The gene expressions of VEGF in mononuclear cells of bone marrow from 7 cases of aplastic anemia and 12 normal controls were detected by semi-quantitative reverse transcriptase polymerase chain reaction(RT-PCR). The expressions of VEGF in bone marrow from 20 cases of aplastic anemia and 20 normal controls were also determined by immunohistochemistry assay. The results showed that expression of VEGF mRNA was found in2 out of7 ( %) bone marrow of patients and in 10 out of 12 (%) bone marrow of normal VEGF mRNA in patients with aplastic anemia was significantly lower than that in normal controls ().No patients with aplastic anemia showed immunohistochemical staining of VEGF in bone marrow , while 3 5 out of 20 (25%) normal controls exhibited VEGF positive marrow of aplastic anemia patients contained less VEGF than that of normal persons (). In conclusion , pared with normal controls , VEGF expression decreased significantly in patients with aplastic anemia at gene transcription level and protEin translation level , it may be related to the defect of angiogenesis and thus hematopoiesis in bone marrow of patients with aplastic anemia.
