Abstract
Part I The expression and meaning of miR-218 and Robo1 in human glioma tissue
Objective:To explore the expression and meaning of miR-218 and Robo1 in human glioma tissue.
Method:The expression levels of miR-218 in 40 cases of different grades of gliomas tissues and 10 control brain tissues were detected by means of qRT- expression levels of Slit2 and Robo1 were evaluated using Western blot and immunocytochemist the relationships between miR-218 and Slit2-Robo1 pathway were analyzed.
Results:Quantitive real-time RT-PCR analysis revealed that miR-218 exression were reduced in Ⅰ,Ⅱ-grade glioma tissues(P<)and markedly reduced in Ⅲ,Ⅳ-grade
glioma tissues(P<) versus normal brain tissuees,that Slit2 is distinctly expressed by normal cerebral neurons,but at very low levels in all-grade glioma tissues, moreover,with the progression of malignant glioma,the expression of Slit2 is gradually decreased or even ,Robo1 is not only expressed in normal brain neurons,but also over-expressed in different grades of gliomas(P< 5).
Conclusion:MiR-218 exression were markedly reduced in all-grade glioma tissues, meanwhile the expression of Slit2 in all-grade glioma tissues were decreased;there was a positive correlation between the expression of Slit2 and miR-218; the expression of Robo1 in all-grade glioma tissues were markedly increased, there was a negative correla tion between the expression of Robo1 and miR-218.
