摘要:目的:优化免疫磁珠分选前列腺肿瘤细胞的方法,提高阳性细胞得率。方法:对比静
置孵育、连续旋转孵育磁珠对分选后细胞得率的影响,并比较分离柱的大小对细胞得率的影
响。结果:连续旋转法孵育磁珠后,分选细胞得率显著升高。分离柱大小与分选后细胞得率
无显著相关。结论:免疫磁珠分选前列腺癌细胞的细胞得率与磁珠和细胞的孵育方式有关。

Optimization of the isolation of prostate cancer cells with

Abstract: Objective: to optimize the isolation of prostate cancer cells with ic cell sorting
for increasing the sorted cell yields. Methods: the cell yields that sorted by quiescent standing
incubation method and continous rotating incubation method pared. furthermore, the cell
yields that sorted by DS sorting columns and MS sorting columns were pared. Results:
continuous rotating incubattion method can increase the cell yields. No significant relationships
were observed between cell yields and the size of sorting : the cell yields of
isolation of prostate cancer cells with ic cell sorting are related to incubation methods.

0 引言
免疫磁珠分选技术是细胞研究中一项常用技术,具有效率高、速度快、分选纯度高的特
点。但是制造商提供的使用方法往往是以血液细胞作为预分选细胞,并无分选其他类型细胞
的介绍。本文将介绍免疫磁珠阳性分选前列腺癌细胞的方法的优化。血细胞体积小,为悬浮
细胞,常常应用常规方法即可得到较好的分选效果。而前列腺癌细胞体积较大,容易沉积,
