黄连药用成分的提取及含量测定
摘要:黄连又名为川连、川黄连、姜制黄连、姜连、萸连等。本品为毛茛科植物黄连Coptis chinensis Franch、或云连Coptis teeta Wall以及三角叶黄连Coptis deltoidea Hsiao的干燥根茎。
其主要功能为清热,燥湿,泻火,解毒。用于湿热,呕吐吞酸,高热神昏,心烦不寐,心火亢盛;血热,目赤,消渴,牙痛;外治湿疹,湿疮,耳道流脓;酒黄连善清上焦火热,用于目赤,口疮;姜黄连清胃和胃止呕,用于寒热互结,湿热中阻,痞满呕吐;萸黄连用于舒肝和胃止呕。
本实验以含量较高的活性成分盐酸小檗碱为评判指标,先对提取溶剂和提取方法进行筛选,然后以溶剂浓度、提取时间和溶剂加量为考察因素,进行用正交设计实验对黄连的提取工艺条件进行优化。结果表明,从黄连中提取盐酸小檗碱,用乙醇作为溶剂要优于甲醇、丙酮,以热回流法提取优于超声提取法。正交设计实验表明最佳提取工艺条件为3g黄连生药粉用150ml的80%乙醇提取3h。
本实验通过对所筛选出的最佳工艺条件进行了验证,表明了筛选出的最佳工艺条件具有一定的可行性,并且具有较高的稳定性,而且所提取的有效成分含量也比其他方法高。

Abstract:Huanglian is named chuanlian ,chuanhuanglian,jiangzhihuanglian,jianglian,yulian and so Branch of huanglian is Ranunculaceae Coptis chinensis Franch or Coptis teeta Wall and Coptisdeltoidea. The major function of huanglian is clearing heart,eliminating dampness discharging fire for muggy,sickness and acid regurgitation a,fuss andsleepness,heart-fire,hyperactivity;blood-heart,hot-eyes,drinking and urine,toothache;it also used for eczema,auditory meatus suppurate;jiuhuanglian is good at clearing upper-heater fiery ,used for hot-eyes,,canker;jianghuanglian is used for clearing stomache and controlling nausea and vomiting,besides,chills and fever interties;yuhuanglian is used for relaxing liver, controlling nausea and vomiting .
In this experiment with higher levels of the active ingredient of berberine hydrochloride for evaluation indicators, First screening the extraction solvent and extraction methods, And then with the dissolvent concentration, dissolvent volume and extraction time for the study of factors to optimized extraction conditions of huanglian by orthogonal test.
