α干扰素对人肺腺癌A549细胞生物学行为的影响

【摘要】目的:研究IFNα对人肺腺癌A549细胞株克隆形成、迁移和侵袭能力以及细胞凋亡的影响。方法:用不同浓度(100、500、1 000、2 000 U·ml-1)IFNα处理人肺腺癌A549细胞(实验组),以未经药物处理细胞作对照(对照组),平板克隆形成实验观察两组细胞克隆形成能力的差异,Transwell小室实验观察两组细胞迁移和侵袭能力的差异,流式细胞仪观察两组细胞凋亡水平的差异。结果:IFNα能够明显降低人肺腺癌A549细胞克隆形成、迁移和侵袭能力,诱导细胞的凋亡,而且在一定范围内与药物浓度成正相关(P<)。结论:IFNα在非小细胞肺癌临床治疗中可能有潜在价值。

[Abstract] Objective:To study the influence of interferonα(IFNα) on the colony formation,migration, invasion and apoptosis of human lung adenocarcinoma A549 cell :Human lung adenocarcinoma A549 cells were treated with a concentration of 100,500,1 000,2 000 U
·ml-1IFNα, ability of cell clone formation was detected by the plate colony forming cell migration and invasion ability were observed in Transwell apoptosis was analyzed by flow :The ability of cell colony formation,migration and invasion were significantly α coulde induce cell apoptosis in a concentrationdependent manner(P<).Conclusions:IFNα may have potential value in nonsmall cell lung cancer treatment.
