阿司匹林对人γδT细胞杀伤消化系统肿瘤细胞的影响

【摘要】目的: 探讨阿司匹林对人γδT细胞杀伤消化道肿瘤细胞株的影响。方法: 用异戊烯焦磷酸法体外扩增人外周血γδT细胞。用不同浓度的阿司匹林诱导γδT细胞和消化道肿瘤SGC7901、 SW1990、 SW480、 SW1116、 LOVO细胞株, 用乳酸脱氢酶法测定γδT细胞的杀伤活性, 用流式细胞术(FCM)检测阿司匹林诱导前后的γδT细胞和SGC7901、 SW1990、 SW480、 SW1116和LOVO细胞凋亡百分率。结果: γδT细胞培养10 %%。~ mmol/L诱导24 h后的γδT细胞对5种肿瘤细胞的杀伤活性最高, mmol/L时杀伤活性呈下降趋势; SGC7901、 SW1990、 SW480、 SW1116和LOVO细胞经不同浓度的阿司匹林诱导24 mmol/L时γδT细胞对SW480、 SW1116和LOVO细胞株的杀伤活性略有增强外, 其他组与对照组比较无明显变化。 mmol/L阿司匹林对
γδT细胞诱导24 h的凋亡率(%)明显高于SGC7901、 SW1990、 SW480、 SW1116和LOVO细胞株(%、 %、 %、 %和%)。结论: 阿司匹林在临床常规使用的药物浓度时可增强γδT细胞杀伤肿瘤细胞作用, 超过这一浓度可明显抑制γδT细胞的增殖能力和杀伤活性及增加γδT细胞的凋亡率, 而对SGC7901、 SW1990、 SW480、 SW1116和LOVO细胞株作用不明显。

[Abstract] AIM: To explore the effect of aspirin on human’s γδT cells killing digestive system tumor cell lines. METHODS: Use the isopentenyl pyrophosphate method to amplify human peripheral blood γδT cells in vitro. Various concentrations of aspirin were used to induce γδT cells and digestive system tumor cells SGC7901, SW1990, SW480, SW1116, LOVO cells lines, LDH assays was used to measure the cytotoxic activity of γδT cells, flow cytometry analysis the apoptosis percentage of before and after induced γδT cells and SGC7901, SW1990, SW480, SW1116, LOVO cell lines. RESULTS: γδT cells were cultivated for ten days which proliferation ratio increase from % to %. When γδT cells induced via aspirin concentrations rang from mmol/L to mmol/L which cytotoxic activity on the five kinds of tumor cell lines was the highest, if aspirin
