天然五倍体泥鳅线粒体基因组全序列分析#

摘要:采用 PCR 产物直接测序法,对天然五倍体泥鳅的线粒体基因组全序列进行了测定,
并对其基因组特点进行了分析。结果显示,五倍体泥鳅的线粒体基因组为双链环状 DNA 分
子,全长 16,643bp,共编码 37 个基因,包括 13 个蛋白编码基因,22 个 tRNA 基因,2 个 rRNA,
以及两个非编码区(控制区和轻链复制起始区)。除 ND6 基因和 8 个 tRNAs 基因外,其它
基因均位于重链。重链的碱基组成为 A %,T %,C %, G %。13 个蛋白
编码基因中,除 COX1 以 GTG 为起始密码子外,其它 12 个均以 ATG 为起始密码子,但终
止密码子的差异较大,7 个基因以 TAA 或 TAG(ND5)为终止密码子,而其它五个基因(ND2,
COX3,ND3,ND4 和 Cytb)却以 T-- or TA-为终止密码子。13 个蛋白编码基因间,有 3
个区域存在碱基共用,包括 ATPase 8 和 ATPase 6 共用 10 bp,ND4L 与 ND4 共用 7 bp,
ND5 与 ND6 共用 4 bp。控制区位于 tRNAPro 和 tRNAPhe 基因之间,长 918bp,控制区
包含了脊椎动物共有的保守区域,包括 TAS,CSB1-3 和 CSB D-F。本研究结果表明,五
倍体泥鳅具备典型的脊椎动物线粒体基因组结构特征。

Abstract: plete mitochondrial genome of the natural pentaploid loach Misgurnus
anguillicaudatus is a circular molecule of 16,643 bp in size, containing 13 protein-coding genes,
22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and two main noncoding
regions (the control region and the origin of the light strand replication) . Most of the genes are
encoded on the heavy strand, except for ND6 and eight overall position of
the H strand is A %,T %,C %, G %. 12 of the 13 protein-coding genes
initiated with the traditional ATG start codon except for COX1, which begins with GTG
, the termination codons of 13 protein-coding genes include seven TAA
and one TAG (ND5), whereas other five genes (ND2, COX3, ND3, ND4, and Cytb) have the
