Podoplanin在恶性肿瘤中表达的研究进展.docPodoplanin 在恶性肿瘤中表达的研究进展[ 摘要] 近年来随着肿瘤的发病率不断上升, 肿瘤的早期发现就显得尤为重要。 Podoplanin 蛋白是一种作为淋巴管内皮标志物的跨膜唾液酸糖蛋白。本文介绍了 Podoplanin 蛋白的分子结构和生物学功能,回顾了 Podoplanin 在肺癌、乳腺癌、食管癌、骨肉瘤、口腔癌等多种恶性肿瘤中的表达情况。得出 Podoplanin 蛋白作为一种特异性蛋白在多种恶性肿瘤组织中有着显著的表达,并且 Podoplanin 在不同恶性肿瘤中的表达差异对于恶性肿瘤的诊断、治疗及预后有着良好的临床指导意义。[ 关键词] Podoplanin ;恶性肿瘤;标志物[ 中图分类号] [ 文献标识码]A[ 文章编号] 1673-7210 ( 2016 ) 05(b) -0041-04 [Abstract] With the rising incidence of tumors , tumors early detection is particularly important in recent years. Podoplanin protein , asa lymphatic endothelial marker , isa kind of sialic acid glycoteipron across cell membranes. This article introduces the molecular structure and biological function of podoplanin protein , and reviewes the expression of the podoplanin in lung cancer , breast cancer. esophageal cancer , a , oral cancer and other malignant tumors. It is concluded that podoplanin protein serving asa kind of specific proteins ina variety of malignant tumor tissues has obvious expression , and the expression of podoplanin in different malignant tumors has an important clinical significance for the diagnosis , treatment and prognosis of malignant tumors. [Key words] Podoplanin ; Malignant tumor ; Markers Podoplanin 又称为平足蛋白,是一种跨膜唾液酸黏蛋白样糖蛋白,目前已经被证实是一种淋巴管内皮标志物[1] 。在近来的研究中已被证实 Podoplanin 蛋白表达于一些恶性肿瘤(肺癌、恶性脑膜瘤[2] 、恶性黑色素瘤[3] 、宫颈癌及结直肠癌等)的肿瘤组织以及瘤旁组织。随着研究的进展 Podoplanon 现已发展成肺癌、中枢神经系统肿瘤、生殖细胞肿瘤的肿瘤标志物。 Podoplanin 作为一种新的免疫组织化学标志物, 在恶性肿瘤细胞的侵袭、迁移和转移中扮演着重要的角色。血小板聚集和肿瘤侵袭、转移的相关研究是当下研究的热点,而 Podoplanin 作为血小板聚集诱导因子, 在促进血小板聚集过程中起着关键作用[4] , 所以 Podoplanin 和恶性肿瘤侵袭、转移之间的关系成为了目前许多恶性肿瘤研究的切入点。本文就 Podoplanin 在多种恶性肿瘤中的表达情况及相关研究进展做一简要综述。 1 Podoplanin 的结构及生物学功能 Podoplani n 是一种相对分子质量为 38kD的Ⅰ型跨膜唾液酸黏蛋白样糖蛋白, 它包括一个呈单一顺序排列的跨膜区域、胞内部分 C 端的 2 个磷酸化位点和胞外的 6 个氧连接糖基化位点(富含丝氨酸和苏氨酸残基)。这种蛋白在 1996 年由 Wetterwald 等[5] 首次发现并报道。在之后的研究中发现这种蛋白存在于小鼠的肾小球足突细胞细胞膜的表面, 和维持肾小球通透性以及足突细胞突起的形态有关,所以将其命名为 Podoplanin 蛋白[6] 。 Podoplani n 具有诱导血小板聚集的生物学功能, 研究[7] 表明 Thr5 2 部分是 Podoplanin 中的唾液酸化核心,被 ATF2 基因所调控, 在诱导血小板聚集方面起到了关键性作用。目前国内外的研究主要是围绕 Podoplani n 作为淋巴内皮标志物和
