Results Hematopoietic stem cells from CD26 /_ mice were successfully engrafted into the recipient and there were no statistically significance in the chimerism between the group of bone marrow alone and bone marrow plus speen T cells 8 weeks post transplantion ( ±%, p>). Moderate GVHD were determined after transplantation in the group with bone marrow plus spleen donor cells. On day 35 after transplantation, the body weight of mice in /he group of bone marrow alone and bone marrow plus speen T cells decreased % and % ,while the GVHD score were and () respectively. Pathological evaluation showed that there were significant change which consist with GVHD in the mice liver, skin and intestine. Conclusion The CD26 gene knockout bone marrow were engrafted into allogeneic WT mice and caused GVHD,

移植物抗宿主病(Graft versus Host Disease, GVHD)是异基因造血干细胞 移植最严重的并发症,红50%的异基因骨髓移植患者发生GVHDU1。但骨髓移植 目前仍是大多数类型的白血病、骨髓异常增生综合症、免疫缺陷病的唯一治愈方 法,每年有数千患者接受骨髓移植。在经历化疗、清髓、造血干细胞输注后, GVHD成为影响患者生存和生活质量的重要因素(2〕。CD26是一种具有蛋白水解 酶活性的糖蛋白分子,分子量左llOkDa,表达于多种细胞表面-由于具有丝氨 酸蛋白酶活性,CD26也被称为二肽基肽酶IV (DPPIV),能够特异性切割N-末 端的脯氨酸或丙氨酸固。CD269PPR」有两种存在形式:细胞表面分子和可溶性 分子,可溶性CD26广泛存在于血清/血浆、脑脊液、关节液中,跨膜蛋白分子 在众多组织中都有表达。同时,CD26也是一种T细胞分化抗原,主要表达壬在 CD4+CD45RO+记忆性T细胞。T细胞活化以后,CD26的表达明显上调—作为 共刺激分子参与T细胞的信号传导网。本项目利用CD26基因敲除小鼠,研究敲 除CD26分子的造血干细胞在异基因小鼠体内植入的情况,建立CD26基因敲除
