1 碱性成纤维细胞生长因子对大鼠骨髓基质细胞体外诱导分化的影响【摘要】目的研究碱性成纤维细胞生长因子( bFGF ) 对骨髓基质细胞( BMSC )体外定向诱导分化时细胞增殖的影响。方法采用全骨髓法分离大鼠双侧股骨 BMSC ,传代后培养液中加成骨诱导剂,分为实验组(培养基中加 的 bFGF ) 和对照组( 培养基中无 bFGF , 其余条件相同)进行培养。通过 MTT 法检测细胞增殖来比较 bFG F 在大鼠 BMSC 增殖中的作用。结果实验组细胞增殖较对照组细胞活跃( )。结论 bFGF 对 BMSC 定向诱导分化具有良好的促进作用。【关键词】碱性成纤维细胞生长因子;骨髓基质细胞;增殖【 Abstract 】 Objective To study the influence of basic fibroblast growth factor ( bFGF ) on the growth of bone marrow stromal cells ( BMSC ). Methods Bone marrow stromal cells ( BMSC ) were isolated from both femur in Wistar rats using 2 all-bone marrow culture method. After subculture of the cells, the cells were induced to osteoblasts by osteogenic inducer. The bFGF were added into culture medium of bFGF group, and control group were cultured without bFGF. By MTT method, the effect of bFGF on proliferation of BMSC was evaluated in vitro. Results The increase of bFGF group was more active than BMSC group ( ). Conclusion bFGF has the ability to promote on bone marrow stromal cells proliferation into osteoblasts. 【 Key words 】 basic fibroblast growth factor;bone marrow stromal cell;proliferation 骨髓基质细胞( bone marrow stromal cells , BMSC ) 具有多向分化潜能, 它可在不同的生物环境和因子的作用下向相应的方向转化, 参与细胞再生。碱性成纤维细胞生长因子( basic fibmblastgrowth factor , bFGF )对体外培养条件下 BMSC 贴附、增殖、分化的影响仍存在争议, 并认为其作用与加入的时间、剂量有关。本实验旨在探讨以 BMSC 作为骨组织工程的种子细胞进行体外定向诱导培养时 bFGF 对其增殖的影响。 3 1 材料与方法 实验材料(1) 实验动物: Wistar 大
