良恶性嗜铬细胞瘤基因表达差异分析
摘要
目的:恶性嗜铬细胞瘤大约占嗜铬细胞瘤的 10%,副神经节瘤中恶
性所占的比例更高些。目前对于嗜铬细胞瘤患者,术后病理学诊断对良
恶性的鉴别尚存争议,临床上将嗜铬细胞瘤转移和复发作为可靠的恶性
诊断标准。本实验旨在筛选良恶性嗜铬细胞瘤表达有差异的基因,并建
立基因学检测实验平台,以期用于在临床水平鉴定良恶性嗜铬细胞瘤。
实验方法: Pubmed 复****近 10 年有关良恶性嗜铬细胞瘤研
究的相关文献 2000 余篇,选出在良恶性肿瘤基因表达有明显差异的 32
个基因,通过实时荧光定量 RT-PCR 的方法,检测在良性嗜铬细胞瘤组
(n=15)及恶性嗜铬细胞瘤组(n=14)中的表达差异。 GeXP (基因
表达谱分析) 的方法定量分析肽处理相关基因在良恶性嗜铬细胞瘤中的
表达。
实验结果:、ZNF496 在两组中表达有显著差异(p<),
能显著区分良恶性嗜铬细胞瘤。
良性组,其中 QPCT、HSPA4L、RNF6、RNF128 四个基因在恶性嗜铬细

胞瘤中的表达显著低于良性嗜铬细胞瘤(p<)。
结论: 1. 肿瘤相关基因 TIMP4、ZNF496 与嗜铬细胞瘤的恶性潜
能关系密切,TIMP4 下调与 ZNF496 上调可作为反映嗜铬细胞瘤生物学
特性的指标。 (基因表达谱分析)技术能够快速、平行地检测多
个肿瘤相关基因的改变,对于探讨嗜铬细胞瘤发生和转移的分子生物学
机制、以及寻找恶性嗜铬细胞瘤特异的分子生物学标志物具有重要意义。

Gene expression profiling of benign and malignant in Chinese
pheochromocytoma patients
Abstract
Objective The prevalence of malignant pheochromocytoma is about 10%,
and is somewhat higher for paraganglioma. Since there are no reliable
discernible morphologic criteria to predict malignancy, malignant
pheochromocytoma has to be diagnosed by the presence of metastatic lesions
to sites where choromaffin tissue is normally absent. In order to explore the
potential biomarkers for monitoring malignant potential, pared the
gene expression patterns between malignant and benign pheochromcotyoma
samples and also provided a platform for the genomic screening.
Methods Fifteen benign and fourteen malignant specimens were from
pheochromocytoma tissue bank. We applied quantitative real-time
polymerase chain reaction(QRT-PCR) to detect expression profiles of 32
tumor metastasis-related genes. These genes were expected to reliably

distinguish benign from malignant tumors by reviewing last 10 years
published article through Pubmed. Besides, quantitative analysis was
carried out, by means of GeXP(Gen
