Sporadiccerebralamyloidangiopathy(CAA)relatestocerebrovas­96-

cularaccumulationofamyloidfibrilsmadeofamyloid-βpeptideAβ401:50to1:-HCl
[1].CAAisveryfrequentinAlzheimer'sdisease(AD)butalsoinnon--existingimmunecomplexes,then
,neutralizedinthesamevolumeof2xPBS4%,

hemorrhagicfeatures(CAA-he)[2]and,inrarecases,withcortico­
sensitiveAβ-relatedCNSvasculitis(CAA-relatedinflammation,CAA-ri)IgG,IgA,orIgMweredetectedusingappropriateantisera,andIgG
[3,4].
PathophysiologicalmechanismsleadingtoCAA-heandCAA-riarefromuncoatedwellsweresubtractedinordertoretainsignalsspecific
-AβantibodiesofIgGclasswereshowntoin­foranti-,IgG2detectiondidnotyieldmeasurableresults.
creaseintheCSFduringCAA-ri[5,6],whichsuggestsanautoimmuneExperimentaldilutioncurveswerefittedtosigmoidmodels[16]that
,ADsubjectstreatedbyinfusionsofalloweddefining3parameters:
monoclonalanti-Aβantibodiesdevelopeddose-dependenthemorrhagicexcess,reflectingtheamountofantigenbindingsites,hencethedi­
andvasculitisfeaturestermedamyloid-relatedimagingabnormalitiesversityofepitoperecognition;,correspondingtothedilution
(ARIAs)[7].Anti-Aβantibodiesbelongtothenaturalautoantibodyathalf-maximumsignal,dependingonconcentrationandavidityof
repertoire,asothernaturalauto-antibodiesinvolvedinneurodegener­polyclonalantibodies;-maximum,
ativedisorders[8–11].GrossserumanalysesinADledtoinconsistentvaryingwithcooperativityphenomenabetweendistinctantibody
conclusions[12],whereasmorerefinedanalysessuchasanti-
subclasses,discriminatebetweenatypical(focal)andclassicalformsoflinearizationprocedure[17].Thesigmoidmodelingofexperimental
AD[13].Toourknowledge,thereisnopublisheddataregardingserumcurvesshowedexcellentgoodness-of-fit(meanR2=),andinternal
anti-Aβantibodiesinspontaneous(non-AD)CAA-riandCAA-%forallfour
Thiscase-controlprospectivestudyenrolled105participants:46parameters.
CAA-heinpatientsfulfillingthemodifiedBostoncriteriaforprobableorTable1presentstheanti-Aβserologicparametersindependently
definiteCAA[14];18CAA-riinpatientsfulfillingthecriteriafornon-associatedwithCAA,CAA-heandCAA-ri,ascomparedtocontrols,ac­
invasivediagnosisofCAA-ri[15];­
-,CAA-he-,and
-rimodelsconsideringindividualanti-Aβprofiles.
ForELISAanalysesofserumanti-Aβantibodiesdilutioncurves,driedAnalysesofbloodanti-Aβantibodiesdemonstratecomplexserolog­
syntheticAβ1–40andAβ1–42aliquotsweredissolvedin10μLDMSO,icalprofilesinCAA,withdistinctivefeaturesinCAA-heandCAA-ri,and
sonicatedtoyieldmonomericforms,and,forfibrillarAβ1–42(f-Aβ42)andsuggeststheexistenceofdefinedcirculatinganti-Aβantibodyspecies

Aβ1–40(f-Aβ40)respectively,-Aβantibody

dilutedto15μg/,160mMmanifestations,assuggestedinexperimentalmousemodels[18,19],and

Inconclusion,thiscorrelativestudydemonstratesdistinctserum
anti-AβantibodypatternsinCAAandresultinghemorrhagicandin­

shouldelucidatethetriggeringroleofanti-Aβantibodiesinspontaneous
orimmunotherapy-inducedCAAmanifestations,andprovideappro­
priatebiomarkersoftheseconditions.
