摘要目恤细胞周期失控是许多肿瘤发生、发展的重要原因, Rb通路是细胞周期G卜s期中相对独立的一条分子信息传
导途径,与多种肿瘤的发生和发展有关。该通路涉及到 P16/cyclin DI/pRb三种基因产物。P16、cyclin D1、pRb 蛋白在许多肿瘤中都存在异常表达。理论上讲,当Rb通路调节环被打破时,p16、cyclinDl、pRb蛋白间一定会有相关联的变化,促使细胞损伤修复,凋亡或癌变。目前认为,神经母细胞瘤(NB)和神经节细胞瘤(GN),体现了神经脊细胞逐渐分化的过程。神经母细胞瘤(NB) 细胞转化与细胞分化障碍是NB细胞多种癌基因突变与多步骤表达调控发生改变的结果。因此,本文重点研究调节细胞周期G1/S调控点的Rb通路在神经母细胞瘤和神经节细胞瘤的发生和转化中是否起到相关的作用。
f方法: 采用免疫组化s—P通用型二步法,检测神经节细胞瘤(39例)及神经母细胞瘤(41例)p16、cyclinDl、pRb
蛋白表达情况。

结果: 经SPSS软件包统计学处理分析,在神经母细胞瘤组和神经节细胞瘤组间p16、pRb蛋白的异常表达率差异具有显著性(p<,x 2检验),cyclinDl蛋白的异常表达率差异具有显著性(p<,X 2检验)。经相关性检验在神经节细胞瘤及神经母细胞瘤中,p16、pRb 和cyclinDl蛋白表达间均无相关性。经x 2检验在神经母细胞瘤中,p16、cyclinDl、pRb蛋白表达与年龄,分期, 原发部位间均无差异显著性。
结论: 、pRb蛋白与神经节细胞瘤肿瘤演变和神经母细胞瘤恶性转化密切相关2:cyclinDl过表达是GN, NB形成中常见的分子水平改变,其过度表达程度可能反映了肿瘤细胞增殖状况。3:P16,pRb蛋白缺失和 cyclinDl过表达三者可单独或共同参与GN,NB的形成。4。 P16,pRb,cyclinDl蛋白的表达与神经母细胞瘤发病年龄,分期,发生部位无关。可能是因为NB细胞在G1期
缺乏细胞周期规律。,.、
