慢性间歇低氧对幼鼠海马神经细胞凋亡及caspase-12表达的影响

【摘要】目的:探讨慢性间歇低氧对幼鼠海马神经细胞凋亡及caspase-12表达的影响。方法:取体重为80~100 g健康雄性SPF级SD幼鼠(3~4周龄)40只,随机分为间歇低氧2周(2IH)组、4周(4IH)组,空气对照2周(2C)组、4周(4C)组,每组10只。以间歇低氧舱建立慢性间歇低氧幼鼠模型。采用TUNEL法检测海马神经细胞凋亡,RT-PCR检测海马区caspase-12 mRNA表达,免疫组织化学染色检测海马区caspase-12蛋白的表达。结果:间歇低氧组凋亡指数(AI)、caspase-12 mRNA及其蛋白表达均高于对照组(均P <);4IH的AI,caspase-12 mRNA及其蛋白表达均高于2IH(均P <)。结论:慢性间歇低氧幼鼠海马神经细胞caspase-12 mRNA及其蛋白表达随间歇低氧时间延长而增加,内质网应激参与慢性间歇低氧幼鼠海马神经细胞凋亡过程。
