【摘要】目的应用蛋白质组学技术,研究***滥用人员与正常人之间血浆蛋白双向
电泳(2-DE)图谱差异,并对蛋白质凝胶图像分析方法进行了优化设计。方法利用固相
pH 梯度等电聚焦(IPG)为第一向,SDS-PAGE垂直电泳为第二向,对血浆样本进行双向电泳。

应用专业图像分析软件 ImageMaster 2D Platinum 分组分析比较电泳图谱,找出具有
稳定差异的蛋白点。结果通过对两组蛋白凝胶图像的分析,平均每张凝胶总蛋白点数为
350±30 个,完全表达差异的点 1 个,实验组表达量上调的点 3 个,表达量下调的点 2 个。
这表明***的滥用可引起人体血浆蛋白的表达发生改变。结论***成瘾者的血浆蛋
白存在特异性蛋白质组改变。该结果为深入研究依据蛋白质组学相关技术来开发可行性治疗
***成瘾病症方案的手段提供了线索。

Proteomic Analysis of Plasma Proteins from Heroin-abuse
Individuals

[ABSTRACT] OBJECTIVE Using proteome technique, to study the differential expression
by 2-DE image analysis of plasma proteins between heroin-abuse individuals and normal men. In
addition, the study applied an optimized procedure to analyze the 2-DE gel images. METHODS
Immobilized pH gradient(IPG) isoelectric focusing electrophoresis of the extracted proteins was
performed to obtain the electrophoretogram of the first dimension, with the second dimension

obtained by vertical SDS-PAGE. The electrophoretograms were analyzed with ImageMaster 2D
Platinum professional software, and the stable pairs of protein spots with differential
expression were obtained. RESULTS On average, approximately 350 protein spots could be
detected massie brilliant blue staining gels respectively, and 1 of these spots was
exclusively shown on the protein maps. Totaled up to 5 spots exhibited quantitative changes. The
result proved that the abuse of heroin could cause a proteomic variation of human plasma proteins.
CONCLUSION Specific changes were found in plasma proteins of heroin-abuse individuals.
The result provides an insight to explore a new feasible treatment for heroin abuse and addiction,
and which might contribute to further study of related researches in proteome technical platform.
