SMAF1基因的异位过表达对脂肪细胞3T3L1增殖和凋亡的影响 (作者:___________单位: ___________邮编: ___________) 作者:韩威段佳慧吴民泸林泽超刘彦华 【摘要】目的观察一脂肪细胞特异性表达的新基因小脂肪细胞因子1(SMAF1)在小鼠脂肪前体细胞3T3L1中的异位表达对脂肪细胞增殖和凋亡的影响。方法构建小鼠SMAF1真核表达载体并稳定转染3T3L1脂肪前体细胞以获得永久异位表达SMAF1基因的细胞株,使用Western blot证实SMAF1基因的表达。体外培养SMAF13T3L1脂肪细胞,以MTT法检测SMAF13T3L1脂肪细胞的增殖;用流式细胞仪对SMAF13T3L1脂肪细胞进行凋亡分析。结果与对照相比,SMAF13T3L1脂肪细胞增殖明显降低,差异有统计学意义();而SMAF13T3L1脂肪细胞凋亡分析结果与对照无显著性差异。结论 SMAF1基因在3T3L1中的异位性表达,对3T3L1前体脂肪细胞生长增殖有显著的抑制作用,而对细胞凋亡无影响,提示SMAF1在脂肪细胞增殖分化中发挥一定的作用。 【关键词】 SMAF1; 脂肪细胞; 异位表达; 增殖; 凋亡 【Abstract】 Objective To evaluate the effect of a novel adipocyte specific expressed gene small adipocyte factor 1 (SMAF1) on proliferation and apoptosis of preadipcyte 3T3L1. Methods A permanent SMAF1 expression construct was established for stable transfection of 3T3L1 and formation of SMAF1 3T3L1 cell line. After invitro culturing of cells,both MTT method and flow cytometry were used to detect and analyze the proliferation level and apoptosis progress respectively. pared with control groups,SMAF1 3T3L1 cell line showed remarkable reduction in proliferation with statistical significance (),while no notable difference in apoptosis between two cell lines.
