白色念珠菌临床株基因分型及耐药性研究

【摘要】目的:了解白色念珠菌的基因分型情况以及不同基因型白色念珠菌的药敏试验结果。方法:应用聚合酶链反应(PCR)扩增白色念珠菌25S核糖体DNA基因内含子区,可转座I型内含子插入片段的数目及大小的不同使得扩增产物不同,根据产物片段大小和数目分型。采用K-B法对不同基因型进行白色念珠菌药敏分析。结果:临床分离的121株白色念珠菌经PCR分为三型:A型83株,B型27株,C型11株。A型为最常见的基因型,B型、C型白色念珠菌对唑类抗真菌药物的耐药率显著高于A型菌株,对其他抗真菌药物的耐药率差异无显著性。结论:不同型别的白色念珠菌耐药谱可能与特定基因型有一定相关性。

Abstract: Objective: To understand the situation of genotype and the result of drug susceptibility of Candida albicans isolated. Methods: Polymerase chain reaction (PCR) designed to span the region that included the site of the transposable group I intron of 25S rDNA of Candida albicans was carried out. The size of the intron and presence of insertion segments in the 25S rDNA were detected by agarose gelelectrophoresis of PCR product. K-B methods was carried out for antifungal susceptibility testing. Results: A total of 121 strains of Candida albicans were divided into three types: 83 genotype A strains, 27 genotype B strains and 11 genotype C strains. genotype A was the main genotype. The results of the antifungal susceptibility testing showed that strains of genotype B and C were significantly more resistant for azole-drugs than strains of genotype A, no satistically difference was found among other antifungal drugs.
