摘要:目的研究庆大霉素慢性损伤后豚鼠前庭终器蛋白激酶C(PKC)活性的变化及其βⅠ,βⅡ,γ亚型的表达,探讨PKC与前庭损伤修复的关系. 方法采用免疫细胞化学法观察椭圆囊斑、球囊斑和壶腹嵴PKCβⅠ,Ⅱ,γ亚型的表达;应用γ-32 P示踪检测前庭终器PKC活性的变化. 结果 PKCβⅠ,Ⅱ,γ亚型在前庭感觉上皮细胞均呈阳性表达,Ⅱ,γ亚型较β1 亚型表达稍弱,,PKCβⅠ,γ在停止注射庆大霉素后1d其表达明显减弱,随后其表达逐渐增加,至1wk其表达已高于对照组,ⅡⅠ,γ表达变化规律基本相似. 结论庆大霉素慢性中毒后豚鼠前庭感觉上皮PKCβⅠ,γ亚型表达及PKC活性均发生变化,,不同亚型可能具有不同的作用.
