摘要:目的:制备出一种高氧亲和力的血红蛋白氧载体。方法:以纯化的猪血红蛋白作为原
料,用 1-(3-二甲氨基丙基)-3-乙基碳二亚胺盐酸盐作为交联剂,并用 N-羟基硫代琥珀酰亚胺
加速聚合反应,将血红蛋白表面的羧基及胺基进行缩合形成肽键,从而使血红蛋白聚合在一
起,形成聚合血红蛋白蛋白——即血红蛋白氧载体。结果:聚合血红蛋白的氧亲和力 P50 =
torr,Hill 系数 n = ,主要分子量分布从 64 kDa - 1300 kDa。结论:以猪血红蛋白为
原料合成出了一种高氧亲和力的血红蛋白氧载体。

oxygen carrier with porcine hemoglobin cross-linked by
EDC

Abstract: A hemoglobin-based oxygen carrier (HBOC) with high oxygen affinity was prepared.
Using the 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide(EDC)as the cross-linker and the
hydroxy-2,5-dioxopyrrolidine-3-sulfonicacid sodium salt (Sulfo-NHS) as the catalyst, the pure
porcine hemoglobin was polymerized and yielding a cross-linked derivative whose molecular
weight distribute from 64 KDa to above 1300 KDa. From the oxygen equilibrium curves and Hill
plots, the oxygen affinity (P50 = torr) and Hill coefficient (n = ) were obtained,the
oxygen affinity was dramatic raised pared with its native protein.
