三种藻类海洋中药多糖的提取及理化性质研究
摘要
本论文以三种藻类海洋中药海藻、昆布、海茜为原料,采用热水回流提取的方法得到海藻粗多糖HZ1,然后采用乙醇分级沉淀的纯化方法对HZ1进行初步的纯化,得到粗褐藻胶HZ2组分和粗褐藻糖胶HZ3组分。采用硫酸苯酚的方法对HZ1进行了总糖含量的测定。采用1-苯基-3-甲基-5-吡唑啉酮高效液相色谱(PMP-HPLC)柱前衍生化方法对HZ1进行单糖组成分析,还用此方法测定了HZ2组分中甘露糖醛酸和古洛糖醛酸的比例以及HZ3组分中岩藻糖的含量。此外,用高效凝胶渗透色谱法测定HZ3分子量。
结果表明,海藻类中药多糖的理化性质既有相似又有不同,推测可能与海藻的种类、提取方法、采收季节、生长年限等有关。本研究为藻类海洋中药材的质量控制和药效物质基础研究提供了初步的实验数据。

Study on Extraction and Physicochemical Properties of three kinds of traditional marine Chinese medicines
Abstract
In this paper, three kinds of traditional marine Chinese medicines(haizao, kelp, Haiqian)were used as raw materials. Algae were extracted with water by heat under reflux method, thus, HZ1 crude harides were acquired, then two fractions crude alginate (HZ2) and crude fucoidan (HZ3) were obtained by fractional precipitation of HZ1 with ethanol. The content of total sugar in HZ1 was determined with phenol sulfate method. The position using precolumn derivatization with 1-phenyl-3-methyl-5-pyrazolone (PMP); the ratio of mannuronic acid and guluronic acid in HZ2 and the content of fucose in HZ3 were also determined with this method. The molecular weight of HZ3 was measured by Gel Permeation Chromatography (GPC).

The results provided data to the quality control and material base of traditional marine Chinese medicines origin from algae.
