干细胞基因 Oct4 蛋白在胃癌细胞内表达意义的研究
目的:
检测干细胞基因 Oct4 在胃癌细胞株 MKN28、SGC7901 和 BGC823 中的表达情况,
干扰高表达 Oct4 的胃癌细胞株中 Oct4 基因表达,研究 Oct4 基因被干扰后胃肿
瘤细胞的恶性侵袭性变化,进而最终探讨 Oct4 基因与胃肿瘤细胞的恶性度关系。
方法:
RT-PCR 技术检测恶性度由高到低的三株胃癌细胞内的 Oct4 mRNA
表达情况,确定 Oct4 mRNA 相对高表达的胃癌细胞株。
Oct4 RNA 干扰片段干扰高表达 Oct4 的胃癌细胞株,并通过荧光定量 PCR
和 Western-bloting 检测 Oct4 基因被干扰后的表达情况。
Transwell 细胞体外侵袭实验检测空白对照组、阴性 siRNA 组以及 Oct4
siRNA 组的胃癌细胞侵袭性变化,重复同样实验三次。
结果:
RT-PCR 结果显示,在三株胃癌细胞中 Oct4 mRNA 表达水平由高到低依
次是 BGC823、SGC-7901、MKN28。
BGC823 细胞的 Oct4 基因进行干扰后,荧光定量 PCR 及 Western-bloting 检
测结果均显示干扰后的 Oct4 表达水平明显下降,与空白对照组比较,差异有统
计学意义(P<)。
细胞体外侵袭实验结果显示,干扰 Oct4 的表达后 BGC823 细胞
(Oct4siRNA 组)穿过膜的数量与空白对照组和阴性 siRNA 组分别比较,均明显
减少,差异均有统计学意义(P<)。
结论:
BGC823、SGC-7901、MKN28,同时检测
Oct4 mRNA 表达水平由高到低依次也是 BGC823、SGC-7901、MKN28,提示 Oct4 基
因在维持肿瘤细胞低分化状态中起重要作用。

Oct4 后的胃癌细胞 BGC823 的侵袭性明显下降,表明 Oct4 基因的表达与
胃癌细胞恶性侵袭能力直接相关,提示 Oct4 在胃癌细胞的侵袭性方面起重要作
用。

Abstract
Objective:
To investigate the significance and expression of stem cell gene Oct4 by
semi-quantitative RT-PCR in three gastric cancer cells MKN28,SGC7901 and
BGC823,and then,to interfer the expression of Oct4 in gastric cancer cell which has a
highest exprssion degree of Oct4 by using a technique called RNA
the Oct4 interfered,to investigate the invasive changes of gastric tumor cells,and then
to explore the relationship between Oct4 gene and the malignant degree of gastric
tumor cells.
Methods:
1. to detect Oct4 mRNA expression from high malignant degree to low among
the three gastric cancer cells lines by semi-quantitative RT-PCR technology,and then,
to find the gastric cancer cells line with high expression degree of Oct4 .
interfer the expression of Oct4 with Oct4 siRNA in the gastric cancer cells
