莱阳农学院颂士学位论文山羊卵母细胞体外成熟和体外受精中细胞核迁移的研究摘要细胞核在生物细胞中占主导的地位,绝大部分遗传物质存在于核中,因而对卵母细胞体外成熟培养和体外受精过程中细胞核迁移的研究是十分必要的。它对于其他jj丕胎工程技术有极大的促进作用。特别是对于核移植中卵母细胞去核方法的研究有极其重要的意义。,对卵母细胞体外成熟和体外受精过程中细胞核的迁移进行了详细地观察。为进一步研究卵母细胞体外成熟和体外受精过程中的各种现象及分子机理提供了基础数据。结果表明: 1、血清、抗生素和卵巢直径对卵母细胞的体外成熟影响不大(差异不显著);切割法得到的可用卵数显著高于抽吸法,差异极显著。 2、山羊卵母细胞的成熟分裂从GVBD现象起,其细胞核即由卵母细胞的近中央位置逐渐移向细胞表面,之后同源染色体分开,排出第一板体。卵母细胞成熟后,随着时问的延长,细胞核又逐渐远离第一极体端。 3、培养开始时绝大部分卵母细胞处于GV期,之后DK期、M I期、A I期、M II期相继出现并达到峰值。 4、应用面积积分法计算得知:体外成熟培养时,0~ 期,;—,;— I期,持续7小时;— I期,持续 ;1 ~22小时为M II期,。 5、雌雄原核形成后逐渐相互靠近,形成合子。关键词:orcein(地衣红) 卵母细胞核迁移体外成熟体外受精面积积分法!!!塑堡!!翌!!!!!羔!!g!!!型!!!!!型!!生!!!!!g垒g!!!!!!坚!!￡!!!望! 垒!!!!型 ThestIldy帅Q粕血al憾M诤伽-0nofGoatdu血g IVMandIVF Absn’act №lcleoIusplays adomina】1trole plaslllmay be itIsnecessary tostudy germjnaI vesicalmjgration IVM and lvF promoIes advancenlent of theotherembryonic engineerjng tcchnology EspecialIy usefulforthestudyofpitted time nucleartr叭sfer.
