锦灯笼抗炎药效物质基础及其作用机制 陈雷 1 何玉静 1 宁立华 1 庞敏 1 邱峰 2 赵烽 1 1 烟台大学药学院 264005 烟台 2 沈阳药科大学中药学院 110016 沈阳摘要:目的: 研究锦灯笼中的活性成分酸浆苦素 A与木犀草素、木犀草苷的协同抗炎作用。方法: 用 LPS 刺激小鼠单核巨噬细胞 RAW 264. 7 释放 TNF- α、 NO 等炎症介质,评价药物对巨噬细胞释放炎症介质的抑制作用。 ELISA 法检测 TNF- α的含量、 Griess 法检测 NO 的含量, 同时以 MTT 法评价细胞毒性。 Western blot 法检测 iNOS 及内参蛋白β-actin 水平。结果: 酸浆苦素 A与木犀草素、木犀草苷联合用药抑制 LPS 诱导 RAW 264. 7 释放 TNF- α、 NO ,有明显的协同作用。 Western blot 结果表明酸浆苦素 A 与木犀草素、木犀草苷联合用药能够显著抑制 iNOS 蛋白水平。结论: 酸浆苦素 A与木犀草素、木犀草苷联合用药具有协同作用,可以为锦灯笼及相关制剂的临床应用提供一定的理论依据。关键词: 酸浆苦素 A ;木犀草素;木犀草苷;协同抗炎作用 Abstract: Objective: To study the synergistic anti-i nflammatory effects of physalin A and luteolin or luteoloside. Methods: Macrophages RAW was induced by LPS to release TNF- α and NO, ELISA was used to detect TNF- α content, and Griess assay was used to test NO, while cytot oxicity was tested by MTT method. Western blot was used for detection of iNOS and β-actin protein expression. Results: Physalin A exerted synergistic anti-inflammatory effects with luteolin or luteoloside. Western blot results showed bination of physalin A and luteolin, physalin A and luteoloside can significantly i nhibit iNOS protein expression. Conclusion: The synergistic anti-inflammatory effects of Phys alin A and luteolin (or luteoloside) may provide theoretical basis of related preparations for their clinical application.
