　　1 材料与方法
　　 淋球菌菌株
　　淋球菌株ST2951用WGC淋球菌专用培养基培养(购自中国科学院武汉生物制品研究所)。
　　 外周血中性粒细胞的分离
　　6名健康成年男性分别抽取2 mL肝素抗凝静脉血，取抗凝血与Hank液混匀后小心加入到人中性粒细胞分离液中(天津灏洋生物制品科技有限公司，LTS1087)，18℃、2000 r/min离心20 min，待离心管由上至下分4层;吸取第3层(中性粒细胞)加入到含Hank液15 mL的试管中，充分混匀，2000 r/min离心10 min，弃上清，沉淀反复洗涤2次，再用含10%胎牛血清DMEM培养液重新悬浮细胞(1

　　 中性粒细胞淋球菌感染模型的建立
　　取出培养板，A组每孔加入含10%胎牛血清DMEM 培养液150μL，B组每孔滴入淋球菌悬液150μL;加样完毕后继续孵育，按0、3、8、12 h时间点在培养孔中吸出培养液作相应检测，每组设3个复孔。
　　 实时荧光定量PCR检测iNOS mRNA含量
　　用Trizol溶液(美国GIBCO公司)提取中性粒细胞总RNA，经紫外分光光度计检测符合纯度后逆转录为cDNA，再进行实时荧光定量PCR检测。根据GeneBank序列设计iNOS、βactin引物，序列见表1。反应条件：94℃预变性3 min，94℃变性5 s，60℃退火10 s，72℃ 延伸20 s，共40个循环。应用SYBR GreenⅠ荧光染料(美国Biotium公司)技术行实时定量PCR反应，获取各组标本的标准曲线，计算机分析Ct值(每个反应管内荧光强度达到系统认为有目的DNA合成时的循环数，以阴性对照作参考)。表1 引物序列
