mi RNA- 126 可靶向抑制 A549 细胞迁移和侵袭能力作用研究* 孙艳芹 1 、刘建强 2 【摘要】目的: 研究 microRNA-126( miR - 126 ) 对非小细胞肺癌( NSCLC ) 细胞迁移和侵袭能力的影响。方法: 利用脂质体介导法将构建好的 miR-126 过表达质粒转染至 NSCLC 细胞株 A549 细胞( A549/ miR-126 组), 并设空质粒转染组( A549/mock 组) 和空白对照组( A54 9 组)。利用 RT-PCR 技术检测三组细胞中 miR-126 及 EGFL7 蛋白表达水平,采用细胞划痕实验观察细胞迁移能力差异和, Transwell 小室法分析三组细胞侵袭能力的差异。结果: RT-PCR 检测发现, 与另两组细胞相比, A549/ miR-126 组细胞中 EGFL7 水平较高, 差异有统计学意义(P< ); 细胞划痕实验发现, 与另两组相比, A549/ miR-126 组细胞迁移距离较小, 抑制率较高, 差异有统计学意义(P< ); Transwell 小室实验发现, A549/m iR-12 6 组 24h 侵袭细胞数分别为(2 ±2 .322 ), 36h 侵袭细胞数为(29 .2± )个, A549/m ock 组为( ± )个, 差异有统计学意义(P< )。结论: miR-126 可上调 EGFL7 的表达, 并可能抑制了 A549 细胞的迁移和侵袭能力。【关键词】非小细胞肺癌( NSCLC ); EGFL7 ; miR-126 miR-126 may inhibit A549 cell migration and invasiveness by targeted EGFL7 Sun Yanqin 1, Liu Jianqiang 2 [ Abstract ] Objective: To study the impact of miR-126 on A549 cell migration and invasiveness.
