摘要:目的:探讨 RNA 干扰技术沉默 B7-H1 基因对肝癌细胞 HCCLM3 生长侵袭及体内成 瘤能力变化的影响。方法:通过慢病毒载体将 B7-H1 小干扰 RNA 片段稳定转染入 HCCLM3 细胞。流式细胞仪检测 sh-RNA-B7-H1 组(感染 B7-H1 慢病毒载体组)、shRNA 对照组(感染 阴性对照慢病毒载体组)K-8 法检测转染后 HCCLM3 细胞生长情况; Transwell 实验检测转染后 HCCLM3 细胞运动侵袭能力;裸鼠皮下接种检测转染后 HCCLM3 细胞体内成瘤及移植瘤生长能力。结果:慢病毒载体干扰 RNA 可以明显降低 sh-RNA-B7-H1 组 HCCLM3 细胞的体外侵袭和运动能力(P<),K8 法检测细胞周期 及体外生长能力发现各组相比差异无显著性(P>)。裸鼠体内实验发现 sh-RNA-B7-H1 组 HCCLM3 细胞的成瘤能力及移植瘤的生长能力显著低于对照组(P<)。结论:本研究通 过 RNA 干扰技术证实下调 B7-H1 表达可直接抑制肝癌细胞的体内生长及体外运动侵袭能 力。 关键词:肝细胞癌;复发转移;B7-H1;RNA 干扰。 中图分类号: 20 Biological effects of costimulatory molecule B7-H1 on human hepatocellular carcinoma GAO Qiang, WANG Xiaoying (Liver Cancer Institute and Zhongshan Hospital, Fudan University, Shanghai, 200032) 25 30 35 40 Abstract: Objective To investigate the effects of B7-H1 expression on the in vitro proliferatin and invasion as well as in vivo tumor growth of human hepatocellular carcinoma (HCC) cell HCCLM3.
