OAS-1基因SNP rs2660与慢性HBV感染者自发性HBeAg血清转换的关系

【摘要】【目的】探讨寡聚腺苷酸合成酶基因-1(OAS-1)位点 rs2660的单核苷酸多态性(SNP)与慢性乙型肝炎病毒(HBV)感染者自发性HBeAg血清转换的关系。【方法】收集126例慢性HBV感染者(HBeAg阳性组58例,HBeAb阳性组68例)和无HBV感染者(对照组72例)的外周血,进行DNA提取和扩增,再利用竞争分化聚合酶链反应(CD-PCR)进行扩增,并结合酶免疫法显色以检测该SNP的基因型,两组间基因型比例或等位基因频率采用?字2检验和优势比(OR)计算。【结果】在HBeAg阳性组SNP rs2660基因型GG + %(17/58)、%(19/116),%(32/68)%(39/136),%(38/72)%(44/144);HBeAg阳性组与HBeAb阳性组或对照组之间的差异均存在统计学意义(基因型:P = ;等位基因:P = ),而HBeAb阳性组与对照组之间的差异无统计学意义(基因型:P = ;等位基因:P = )。【结论】 SNP rs2660等位基因G可能有助于慢性HBV感染者发生自发性HBeAg血清转换,其基因型检测对于干扰素治疗慢性HBV感染者可能有一定的疗效预测价值。

Abstract: 【Objective】 The aim of this study was to investigate the relationship between the single nucleotide polymorphism (SNP) rs2660 of oligoadenylate synthetase gene-1 (OAS-1) and spontaneous HBeAg seroconversion on patients with chron
ic hepatitis B virus (HBV) infection. 【Methods】 Blood samples were collected from 128 patients with chronic HBV infection (58 HBeAg positive, 68 HBeAb positive), and 72 normal control cases without HBV infection. Chromosomal DNA was extracted and OAS-1 gene was amplified. SNP genotyping was performed with the enzyme immunoassay (EIA) after petitively differentiated polymerase chain reaction (CD-PCR). Odds ratio and chi-square test were applied in statistical analysis. 【Results】 In HBeAg positive group, frequencies of genotype GG plus GA and allele G were % (17/58) and % (19/116) on SNP rs2660, respectively.
