人参皂苷Rg1对脑挫伤后大鼠脑内胰岛素样生长因子1表达的影响
【摘要】探讨人参皂苷Rg1对脑组织挫伤后大鼠脑中胰岛素样生长因子1(insulinlike growth factor1, IGF1)表达的影响。方法:Wistar大鼠26只,分为正常对照组(2只),脑挫伤模型组(8只)及人参皂苷Rg1治疗组(16只),在损伤后不同时间点(损伤后6 h、12 h、2 d及6 d)分别处死大鼠,使用免疫组化方法观察并检测脑中IGF1的表达。结果:与对照组相比,脑挫伤模型组损伤后各时间点脑组织中IGF1的表达有所升高(P<);与模型组比较,人参皂苷Rg1治疗组IGF1的表达明显升高(P<)。结论:人参皂苷Rg1可能通过促进脑挫伤后脑组织IGF1的表达,促进脑组织的功能恢复。

Objective: To study the effects of ginsenoside Rg1 on the expression of insulinlike growth factor1 (IGF1) in the brain of rats after the experimental brain contusion.
Methods: A total of twentysix Wistar rats were randomly divided into normal control group (n=2), untreated group (n=8) and ginsenoside Rg1 group (n=16). Brain injuries were
induced in rats by a mechanical striking device. The brain tissues were extracted at different times after brain injury (6th hour, 12th hour, 2nd day, 6th day), then the expression of IGF1 in brain tissue was examined by immunohistochemical method.
Results: parison with the normal control group, the expression of IGF1 in the brain tissues was increased in the untreated group after the brain contusion (P<). The expression of IGF1 in brain tissues in ginsenoside Rg1 group was significantly increased pared with the untreated group (P<).
Conclusion: Ginsenoside Rg1 enhances the recovery of the contused brain through increasing the expression of IGF1.

人参皂苷Rg1是人参中提取的一种有效成分,具有神经保护和促进神经再生的作用,近年来有研究证实人参皂苷Rg1的这些作用是通过多种脑内因子实现的[13]。胰岛素样生长因子1(insulinlike growth factor
1, IGF1)是脑损伤后一种重要的内生性神经营养因子,近年来有研究发现创伤性脑损伤及脊髓损伤后,IGF1在神经组织中表达增强[4],可能与脑损伤后神经修复和周围神经损伤后变性有关[5]。本实验采用Feeney's法制作大鼠脑挫伤模型[6],对大鼠注射人参皂苷Rg1,观察其对脑内IGF1表达的影响。
1 材料与方法
实验材料成年Wistar大鼠26只,四川大学华西实验动物中心提供,性别不限,体质量200~250 g。随机分为正常对照组(n=2),模型组(n=8),人参皂苷Rg1组(n=16)。人参皂苷Rg1粉剂,昆明同德众源生物科技有限公司提供,批号110704200605,用生理盐水稀释至10 mg/ml。兔抗大鼠IGF1单克隆抗体(稀释度1︰200)、SP免疫组化染色试剂盒,北京中杉金桥生物技术有限公司提供。二氨基联苯胺显色试剂
